چکیده

The photo-oxidation of lysozyme (hen’s egg white), sensitized by methylene blue in 84% acetic acid solution, causes the selective modification of the methionyl residues, which are converted to the sulfoxide. The photo-oxidized protein shows a drastically reduced enzymic activity. The data obtained by comparing the sensitivity to proteolytic enzymes, as. well as the accessibility of disulfide bonds to reducing agents and the optical parameters, for the native and the photo-oxidized lysozyme, allow one to correlate the loss of catalytic efficiency attending photo-oxi.lation with a conformational change of the protein, The segments of the polypeptide chain containing some tryptophyl residues play a major role in such rearrangement. Chemical reduction of the photo-oxidized lysozyme by aqueous 2-mercaptoethanol results in the restoration of the primary and tertiary structure, characteristic of the native lysozyme. A high recovery of enzymic activity is also obtained.

Dye-sensitized photo-oxidation of enzymes.

Heart lipoamide dehydrogenase, liver alcohol dehydrogenase and egg-white lysozyme are photo-oxidized in the presence of various dye sensitizers. The photodynamic process is preceded by the binding between the enzyme and the sensitizers. Among the commonly used dyes, halogenated xanthines and thiazine are effective sensitizers for the photo-inactivation of these three enzymes. Histidine residues...

Lysozyme Photochemistry as a Function of Temperature. The Protective Effect of Nanoparticles on Lysozyme Photostability.

The presence of aromatic residues and their close spatial proximity to disulphide bridges makes hen egg white lysozyme labile to UV excitation. UVB induced photo-oxidation of tryptophan and tyrosine residues leads to photochemical products, such as, kynurenine, N-formylkynurenine and dityrosine and to the disruption of disulphide bridges in proteins. We here report that lysozyme UV induced phot...

Study on Trend of Biodegradability of Phenolic Compounds During Photo-Fenton Advanced Oxidation Process

Phenolic acids constitute a major group of pollutants which are recalcitrant to common biological treatment. In this study synthetic wastewater containing a mixture of p-coumaric and p-hydroxybenzoic acids was evaluated for photo-Fenton pretreatment. The changes in biodegradability (ratio of biochemical oxygen demand to total organic carbon (TOC)) and mineralization (TOC removal) were monitored...

Selective oxidation of sulfides to sulfoxides by a vanadium-based catalyst using 30% hydrogen peroxide

The vanadium-based catalyst, acts as a homogeneous catalyst for the selective oxidation of various kinds of sulfides with 30% aqueous H2O2. Vanadium-based catalyst was successfully used as the oxygen source for the oxidation of sulfides to sulfoxides for the first time. The sulfoxides were obtained in a good way to high yields without any detectable over-oxidation to sulfones under normal condi...

The relationship between Indicators of plasma oxidative stress and photo aging

Introduction: Photo aging is significantly different from chronological aging in both clinical and histological appearances. It has been suggested that oxidative stress generated by ultraviolet radiation (UVR), leads to photo aging over a long time period. This study was conducted to determine the relationship between the indicators of plasma oxidative stress and photo damaged skin in patient...
